Developing differentiated solutions for the next generation of in vivo CAR-T therapies
Tempest Therapeutics is a clinical-stage biotechnology company focused on pioneering a pipeline of next generation in vivo CAR-T cell therapies for autoimmune and oncology indications. Through a dual-target CD19/BCMA CAR structure, as well as a proprietary CD7-targeted mRNA lipid nanoparticle CAR-T delivery platform, the company is expanding the potential of next-generation cell therapy with broader T-cell reach, better intracellular delivery, and scalable repeat dosing. TPST-4003, the company’s lead in vivo CAR-T candidate, combines these technologies to support broad B-cell lineage reset across multiple indications. The company is planning to advance TPST-4003 in investigator-initiated trials for myasthenia gravis (MG) and multiple sclerosis (MS). TPST-2003 is an autologous CD19/BCMA dual-targeting CAR-T therapy in a Phase 1/2a study in patients with relapsed/refractory multiple myeloma and is being developed in China by Tempest’s partner, Novatim Immune Therapeutics (“Novatim”). Additional preclinical programs in the pipeline leverage advanced payloads and create a modular in vivo CAR-T portfolio.
